CJC-1295 without DAC, also called Modified GRF (1-29), is the first 29 amino acids of growth hormone-releasing hormone with four substitutions that protect it from enzyme breakdown. Ipamorelin is a pentapeptide that activates the ghrelin receptor (GHS-R1a). The two act on different receptors in the pituitary, which is why they are so often studied together.
Researchers use the pair to study pulsatile growth hormone release: how GHRH-receptor and ghrelin-receptor signals combine, and how the short-acting no-DAC form mimics natural release pulses compared with long-acting analogues.

DAC (Drug Affinity Complex) is a chemical group that binds albumin in the blood and stretches CJC-1295's half-life to several days. Without it, CJC-1295 is short-acting, lasting around 30 minutes, closer to a natural GHRH pulse.
The pairing is studied for how GHRH-receptor and ghrelin-receptor signalling combine to drive pulsatile growth hormone release.
